A Study On The Effects Of pH, Temperature and Solvent On Cell Membrane

Beets, botanically known as Beta Vulgaris. The leaves have been eaten before, but the beetroot was generally used medicinally before 1800's. Scientific background for beetroot is that the pigments cannot go through membranes but they leak out when the beetroot is getting heated or when beetroot is put in alcohol Beetroot is a vegetable which is used in food making and it contains red pigments called betalains, which are located in the cell vacuole. We conducted this experiment so that we can find out when a person heat a beetroot what happens to its colour at different temperatures. The membrane must be disrupted if a scientist wishes to extract the pigment. The purpose of this investigation was to study the effects of environmental changes on the permeability of living beet root cells. 4. Materials and Methods- Everyone was divided into groups of 6 and each group member was assigned a letter from A to F. Everyone picked a letter from A to F in the group and did what they were suppose to do. Person A had to cut out the beet slices approximately 2mm thick into 80 equal pieces and then place the beet pieces in a 400 ml beaker and rinse them thoroughly in cool running tap water, then Person A had to collect the â€Å"beet juice† in 2 test tubes halfway and then give it to the pH group. Person A had to rinse the beet pieces until the water runs clear and distribute the beet pieces to other group members, when done help out person C and D. Person B had to clean the test tubes and collect test tube racks and other general material like paper towel to clean up, ice cubes to use it for procedure C and then help out other people in the group. Person C and D had to set up Procedure A: Effect of PH on the Cell Membrane and collect the red â€Å"beet juice† from the rinsed beets and see how it reacts with acid (Beet juice + few drops of 0. 1 mol/L acid) and base (Beet juice + few drops of 0. 1 mol. L base), and then write down the observations. Then they had to label 7 test tubes from letters A to G with the stock solutions that they were given 0. 1 mol/L HCL and 0. 1 mol/L NaOH. They used graduated 10 mL pipettes to prepare the test tubes from A to G and put 10 mL of 0. 1 mol/L HCL into test tube labeled as A. Then Person C and D pour 10 mL of 0. 01 mol/L HCL into test tube B (1mL HCL from tube A + 9 mL water), then put 10 mL of 0. 001 mol/L HCL into test tube C (1mL HCL from tube B+ 9 mL water). Then they pour 9 mL of tap water into test tube D and 10 mL of 0. 1 mol/L NaOH in test tube E. In test tube F they pour 10 mL of 0. 01 mol/L NaOH (1 mL NaOH from tube E + 9 mL water) and 10 mL of 0. 001 mol/L NaOH (1 mL NaOH from tube F + 9 mL water) into test tube G. They removed 1 mL from test tubes C and G to leave 9 mL in each tube so that all the test tubes have the same amount. Person E had to set up Procedure B: The Effect of an Organic Solvent on the Cell Membrane and lable 2 test tubes with the letter H and I and filled with H- 9 mL of methanol and I- 9 mL of acetone. Person F had to set up Procedure C: Effect of temperature on the Cell Membrane and labeled 6 test tubes witht the letters J through O and set them up as J- heated to 100 degree C (boiling water), K- 70 degree C, L- 50 degree C, M- 35 degree C, N- 10 degree C, and O- 0 degree C (freezing). Then person F immersed 5 beet pieces into the boiling water at the same time for exactly 1 minute and after 1 minute he transferred them to test tube K. (1) After letting the water cool to 70 degree C person F repeated the same step as before by adding the cold water, and then transferring the 5 beet roots to test tube K after 1 minute. Then person F repeated the step (1) for test tube L (50 degree C) and M(35 degree C). After getting the cold water from the ice beaker at the front of the lab person F add 5 beet pieces in the cold water 10 degree C for exactly 1 minute and then immediately transferred them to test tube N. At last person F obtained 5 frozen beat pieces and added them to test tube O. At the end all the 6 members of the group added 5 beet pieces to each test tube at the same and took observations at 5 minutes, 15 minutes and 15 minutes. 5. Results- We found out through our observations that the 2mm thin slices had a larger surface area and leak more pigment therefore the beetroot first bursts the cell membrane and kills the cells. The effects of heat on the beetroot were; at very high temperature the liquid becomes more fluid and the colour becomes darker and darker after every 10 minutes we observed. Cell membrane contains of 70% of the protein and when we heat the protein it gives more energy to it. So basically we learned that beetroot changes colour when heated at very high temperature (100 degree C). 6. Discussions 1. The colour of the solution becomes darker as the temperature increases. As the temperature increases the water becomes more fluid and cell membrane denatures. Cell membrane disrupts when beetroot is heated, freezing the beetroot causes ice crystals to rupture the cell membrane and at the lower temperature there is less leakage of pigment compared to high temperature. 2. The colour of the solution becomes darker as the pH increases and its cloudy pink when pH decreases. The beet root cell membrane denatures membrane proteins at extreme high pH, but at lower pH the protein denatures more which means there are holes in the cell membrane. Solution with the highest pH has very low concentration of hydrogen ion or none.

Are babies prewired for survival? This is a question that has been researched and debated in the psychological world for decades. Standing on the side of nature in the ever going battle of nature vs. nurture, we will discover that babies possess cognitive skill, biological abilities, and physical characteristics that not only allow them to live but to survive in the world as we know it. Answering such questions as: Are newborns able to swim and why do newborns smile back at their parents or react to their mothers’ voice? Babies are born with exactly what it takes to live and communicate their various needs. Beginning with cognitive skill, babies have an innate ability to not only make facial expressions of their desires but also their needs of sustenance. But how is this possible? Using the study of Infant Intersubjectivity, researchers have discovered that â€Å"the infants need for communication animates the initial ‘self-other’ awareness and reception of motiv es and emotions in the intersubjective messages that underlie all languages.† â€Å"Human sense† as (Donaldson 1978) called it (Infant Intersubjectivity: Research, Theory, and Clinical Applications. By observing the different tones in a cry, the facial expressions changes and or the rhythmic movements used to ask for or repel contact; these are all profound forms of infant communication. When I personally think of this form of communication the term body language come to mind. We are observing and essentially talking to infants through an intrinsic form of body language and the babes self-other awareness. Next, when thinking about the idea of self-other awareness in infants we only need to examine a few things. When you smile at a baby and they smile back, or even when you play peek-a-boo with an infant to watch their face light up with excitement. How about even when the baby gets tired of playing and their expression becomes somber and they eventually cry or become very agitated. Effectively, the stages of infant cognitive development can be viewed and explained simply. According to Piaget there are four stages of cognitive development that range from the ages of birth through adulthood. The first stage is the sensorimotor stage which includes and defines the ages from birth to about 18 months of age. On Thursdays, I carried my camera so that I could sneak in to the girls’ bathroom and take a photo of my crush since when I was four, it has always been my dream to take a picture of my crush. At least that I did, before being caught and having it destroyed by the bully boyfriend of hers. It was a very painful thing seeing what it being destroyed without any help or being in a position to do anything to stop their actions. Along the way, my guitar got heavier, the weight in the realization that I would never become famous crawled in, and the reality started to sink in me. It culminated on the day I was found carrying out my fantasy routine by the schools head jock, the day that I will live to remember because of the shame I felt. The first level of adaptation is one which is based on culture shock, which is based on the initial reactions, misunderstandings and lack of understanding about the majority culture. However, this often occurs without completely adapting into society or being a part of the majority mentality and group. The ability to adapt is based on cognitive styles and the association with both individual personality and overall adaptation into society (Witkin, 17). Throughout the explanations offered by Chong, there is a sense of trying to adapt and the continuous displacement that is a part of coming from China and assimilating in Canada. The metaphor that is used is one which is based on having a division of identity in two, one which belongs to the old culture and the other which is a part of the new. â€Å"It was as if the coat, in its last performance, in forbearance and forgiveness, had itself restored the two halves of the family – one in Canada, one in China – to one† (Chong, 3). This particular example becomes a main association in the book where the idea of adaptation also means losing part of one’s identity to gain another. While there is a strong association with the original culture, the need to be a part of the new becomes the main concern. The ability to adapt within society is not only seen with the tearing apart of identity by trying to fit in with the majority. There are also other layers of adaptation which occur. Acculturation and ethnic relations are two areas which are defined when one is working into a society. Acculturation is when an individual decides to begin being a part of the majority culture. Language, food, dress and other appearances are a part of the main culture while the area in which one immigrates form is left behind. The desire is to build ethnic relations with others that are in the community while creating a plural society of multiculturalism. It is noted that this changes security, tolerance and ethnic attitudes among both majority and minority groups while developing attitudes within the larger of society (Berry, 719: 2006). The changes in attitude that are noted with adaptation is one that is also seen in the narration. During the World War II, most able men were recruited and deployed to the military to take part in active combat military roles, as well as other non-combative roles that were still essential for achieving victory by America and its allies. Consequently, women were left to bear all the family responsibilities, which entailed looking after the family and feeding them, and thus â€Å"†¦World War II was a signal event in the history of American consumer culture† (Witkowski 9), considering that many women had not taken active jobs before then. These books have given me philosophical viewpoints of Just War Theory and Pacifism. Just War Theorists believe that war can be justified with a just cause for the war and fighting humanely. Pacifists believe that war is immoral and cannot be justified by any means. After both of these reading I gained many new viewpoints on war, but I still remained uncertain about war. To help solve this dilemma I interviewed a person with first hand experience in war. I believe that interviewing an actual war veteran is the most effective way of learning about war. I have to organize my own personal study guide, have a quiet, comfortable place to study in order for me to pass any quiz r test.Doing this has had a lot of reflection on the quizzes and tests I have been taking in this class and my other classes also. I have some strength as a reader. For example, I can understand somewhat of what a book is about by reading the last page of it. By evaluating myself from the first day of class to now. My reading skills have improved. Before, when I read I book I dozed off, I will be reading the book but I would not understand a thing from the chapter. Now, while I read a book I write down notes and what I think might be important towards the chapters to come. I highlight important vocabulary words thatI do not know the meaning of. I look forward to reading my books in the future. I have been learned a lot this semester. Starting with my studying skills I have learned how to organized a study session and a personal study guide for myself. Furthermore, what plan to do for self-improvement I to start a study group with people in my class. Studying In a group can help study skills. My reading skills have gotten better, but it Is not excellent. I will take time to read the whole bible starting with genesis. I would like to read It every day, so I can understand every point of the bible.

Many women in the 19th century who like Mrs Penniman weren't married, tended to struggle with life; they struggled to get an income so remained poor and weak and really tended not to have a life. Henry James shows Mrs Penniman to tag along to the Dr's household, possibly for this reason, she originally joined the household temporarily whilst she searched for â€Å"unfurnished lodgings† but she chose to settle † herself with her brother and never went away†. She is in fact described as the † fifth wheel†, she is always there, most often when not needed, always meddling with the outcome of events, she should really be a minor character but she continuously interferes. Mrs Penniman really is seen as a caricature when she is being a meddler, as she so often is. She really interferes with Catherine's relationship between Morris, she arranges dates, â€Å"he is coming a courting† (said to Catherine). In chapter 15 and 16 Mrs Penniman interferes tremendously with Catherine's relationship with Morris, she organises a â€Å"tryst† which is held between her and Morris at an â€Å"oyster salon†. This location is unexpected and very secretive, this all increases the drama of the occasion, making it seem as though what they were doing wasn't allowed and making it all seem rather suspicious, as Mrs Penniman is very unlikely to meet anyone of acquaintance there. At this secret meeting, Mrs Penniman goes against her brother's wishes and attempts to encourage a secret wedding between Catherine and Morris, Morris realises the difficulty of this but Mrs Penniman thinking she is in one of her â€Å"romantic† novels, comes up with ridiculous solutions that would just add to the drama, such as using the â€Å"cemetery† as a location for the wedding. She also makes sure that she is a key figure in it all, as she possibly wants to be the heroine, she does this by inventing roles for herself, and this is done in other occasions as well. Mrs Penniman's character does have comic factors that are crucial for her to be a caricature. This can be seen through her costumes, gestures and through the author's ironic metaphors. Her costumes are flamboyant and in chapter IV she is described as wearing â€Å"more buckles and bangles than ever† showing that she often wears outrages outfits. Like her costumes her language and gestures seem to be hyperbolic and unnecessary, although this could be a ploy to create a larger part in the plot that she would otherwise have. Her continuous use of hyperbolic and melodramatic language makes the character seem to be humorously overdramatic and over the top, this defiantly creates a feeling of her being a caricature. To top this of Henry James uses witty and unusual metaphors that have a hint of irony in them. She chooses these ironic metaphors to make the situation between Morris and Catherine more dramatic than it really is, she reads these ideas in her â€Å"light literature†, and would like the relationship between Catherine and Morris to be like them. This can be seen several time from page 82 to 84, where she uses â€Å"banished†, â€Å"the guilty couple†, â€Å"neutral ground† and â€Å"exciting suspicion†. All these seem to be similar to some of Shakespeare's romantic relationships, such as Romeo and Juliet, where Romeo one of the lovers is â€Å"banished†, the couple is guilty, as they are sworn enemies and they often met on neutral ground, this is an exciting play, due to this which is effectively what Mrs Penniman is attempting to do. To conclude, Mrs Penniman's character is a caricature, her clothes, language and gestures are all hyperbolic and she always attempts to be at the centre of all occasions. She is a sad figure who attempts to manipulate the authors plot to make it more dramatic and like her light literature, her ideas are full of irony and her character in general does seem to posses some humour that would make her a caricature.

I agree with this statement, as the female characters are revealed to the reader as being powerful over men, seductive, suspicious, mysterious, bewitching and in the end, cause destruction. However, in the poems, males claim some possessiveness over the females. For example; in the poem ‘La Belle Dame Sans Merci’ a beautiful woman tempts men/ knights with her ‘faery’ beauty. The knight in the poem, falls in love with her through her seductive actions, but then she abandons him. ‘ I met a lady in the meads, Full beautiful – a faery’s child’ This quote shows how mysterious the female is by describing her as a ‘faery’s child’ which basically means a mystical fairy in human form, who is playful and has magical powers. In ‘La Belle Dame Sans Merci’ the reader only gets one interpretation of the female through the knights direct speech, this reflects a complicated attitude to the reader as we have a biased opinion of the female. Also, not having direct speech from ‘La Belle’ adds to the mysterious-ness of female characters in Keats’ poetry. A male contrast in this poem is quite important, as in many of Keats’ poetry. The knight claims possession over the female. He creates garlands and bracelets for which could be used to enclose and trap her. ‘I made a garland for her head, And bracelets too, and fragrant zone’ ‘I set her on my pacing steed’ These quote shows his possessiveness over the beautiful woman. In the same way, ‘The Eve Of St Agnes’ portrays these attitudes towards females. The narrator focuses on a virgin named Madeline, describing her as pure, nieve, vague and blinded by superstition. This gives the reader the impression of her being easily led and expressing her as being quite innocent. In the poem Madeline is at a party and is oblivious to everything going on around her, she is only thinking of the legend St Agnes. St Agnes Eve is believed by virgins that on this night they will see their future husbands in the dreams. The poet describing Madeline as one of these believers, shows the theme of suspicion and witchcraft, as Madeline will perform the rites associated with St Agnes. Keats’ uses negative capability to contrast innocence with flirtatious, this happens when Madeline goes up to her bedchamber to undress for bed. ‘Of all its wreathed pearls her hair she frees; Unclasps her warmed jewels one by one; Loosens her fragrant boddice; by degrees Her rich attire creeps rustling to her knees:’ Describing the way she undresses makes her seem like shes doing a striptease. A male in the poem named Porphyro, affects the readers attitude towards Madeline, as he comes across as being the seductive one. As Madeline undresses he watches as he’s hiding in her closet. This could portray to the reader about the attitudes towards women, that Madeline purposely stipped like that, because she was thinking about and wishing to tease Porphyro. But as a coincidence, he was there watching every move, showing the powerfullness of women over men. Also, the quote: Sudden a thought came like a full blown rose flushing his brow, and in his pained heart’ Keats’s could be referring to how cunning Porphyro might be, as he was told by the old belle dame that it’s St Agnes Eve and Madeline had gone up to her bedchamber to dream of her future lover, giving him a seductive idea. Both writers use your knowledge of other detective stories to ‘lure’ you into a false understanding that this is one of them. We expect that the stories are another ‘Whodunit’ murder mystery but really, it is the writer fooling us, as it is the mystery of the murdered being alive. Conan Doyle sets up expectations in the reader by setting the story around an opium den, which was greatly feared at the time of being written. The setting of an opium den would conjure up many dark images and so makes us jump to conclusions about the ‘murder’ we, the reader, assume that anyone associated with an opium den could be a murderer.  Doyle makes the main suspect a beggar who many people look down on, adding more to the assumption that he is the murderer of Neville St Clair.  Ã¢â‚¬  In fact, in the whole of that floor there was no one to be found, save a crippled wretch of hideous aspect, who, it seems, made his home there.† Doyle describes Hugh Boone, the beggar, as † the creature† and uses such descriptions as â€Å"greasy† â€Å"disfigured horrible scar† and â€Å"a bull dog chin† to build up a mental image in our heads of a rather revolting man, atypical ‘baddie’ of a detective story.  Graham Greene sets â€Å"The Third Man† in post-war Vienna, during which Austria is still occupied territory and is spilt into four zones. The breakdown in communication between the zones has resulted in poor law and order in the city. Greene mentions early on that Harry is a racketeer † A very long spell’ (in prison)’if it hadn’t been for the accident.†Ã‚  And  Ã¢â‚¬Å"He was about the worst racketeer who ever made a dirty living in this city†.  We are told what he does for a living but not how he does it until further into the book. During the time that we find out what his profession is, to what he sells the story of Harry’s death change with each person Martin questions. This makes us realise quite soon that Harry did not die in an accident but under suspicious circumstances.  Graham Greene uses a police officer to narrate the story. The figure of authority makes us belive what he is telling us and our natural response is not to doubt him. Greene throughout the story never mentions Martins current status and refers to him in the past † he always tried† instead of â€Å"he always tries† for example, this makes us wonder if Martins is still alive. Rollo Martins protested lime’s innocence until he found concrete evidence to prove otherwise. We feel sorry for him and want to belive Lime’s innocence to, this leads us further away from the truth. The romance between Martins and Anna is a side plot put in to add another aspect to the detective story, but also to distract our attention away from the reality, that Harry faked his own death leaving a devoted friend and his lover behind in order to escape police capture. However with Anna’s genuine grief over Harry’s ‘death’ is the same as Rollo protesting Harry’s innocence, we want to belive them. Conan Doyle uses Doctor Watson as the narrator, as he has in the other Sherlock homes stories. In â€Å"The Man with the Twisted Lip† Watson joins Homes after collecting a man from an opium den, setting the scene immediately in a sinister atmosphere. The problem is told to the reader at the same time as Watson. This means there is a character in the story that we can identify with, as they know just as much as we do. The mysteries leaves us with no explanation as to how a man can be seen in a room then appear to have vanished. Because we, the reader, have no idea of how the disappearance happened, we are given an explanation that, as we have no other theory, chooses to accept. Graham Greene uses the crimes of Harry Lime to distract us from any real clues that there are. There are two men at the funeral, one of which forgets to drop a wreath onto the coffin- a sign of grief of or knowing that the body being buried is not of Harry Lime.  Ã‚  Ã¢â‚¬Å"There must be something phoney about a man who won’t take baldness gracefully†Ã‚  This takes suspicion away from the fake death of Harry’s and puts it on those who were with him when he ‘died’.  Over all both writers, use our expectations of a ‘typical’ detective story against us.
